The University of South Carolina is offering licensing opportunities for Novel imidazothiazoles inhibiting indoleamine 2,3-dioxygenase 1 (IDO1) and tryptophan 2,3-dioxygenase (TDO).

Background:

The kynurenine pathway (KP) is involved in the modulation of many biological and physiological processes including immune response and neurotransmission. Overactive KP has been linked to a wide range of diseases and conditions such as cancer, depression, chronic/neuropathic pain, neurodegeneration, HIV immune dysfunction, etc. Regulating PK by inhibiting the activity of IDO1 and TDO is therefore a strategy for the development of medical therapeutics for these diseases.

Invention Description:

This invention consists of the development of dual inhibitors targeting IDO1 and TDO, which catalyze the initial rate-limiting step of the KP. The inhibition of both IDO1 and TDO result in the regulation of the KP, and is therefore an important strategy for the development of medical therapeutics for these diseases.

Potential Applications:

Most of drugs tested in the clinical and preclinical studies on the market today are made up of selective IDO1 inhibitor. These clinical trials have not yielded successful results, most likely because TDO activity is not included in the trials. As selective inhibition of IDO1 may be compensated by TDO activity, the compliment of both in this innovation may help alleviate this problem. This innovation may be used for the treatment of cancer, neurodegenerative diseases such as Parkinson's disease, and HIV immune dysfunction.

Advantages and Benefits:

The synthetic method to these imidazothiazole compounds is not complicated. The reactions do not employ extreme conditions. Scale-up of the reactions is feasible. The costs of manufacturing are low.
